The results demonstrate a profound depletion of plasma-lineage clusters that typically persist after CD19 single-targeting therapy, alongside a reconstruction of the B cell receptor repertoire toward a healthy, naïve-biased state. The most critical clinical finding is that this dual-targeting approach achieves a significantly deeper depletion of pathogenic plasma cell lineages, facilitating a powerful humoral immune reset. However, when critically appraising this study, clinicians must note that the cohort size is extremely small, with only five patients, and the comparison to CD19-only therapy relies on indirect, cross-cohort analysis. Despite these limitations, this paper is highly relevant for rheumatologists and physicians, as it highlights a potential paradigm shift toward curative cellular therapies for treatment-resistant SLE, paving the way for achieving comprehensive and durable immune remission in future clinical practice.
